T lymphocytes are capable of recognizing and eliminating tumors with exquisite specificity through the recognition of mutated tumor proteins, as a consequence of the accumulation of genetic alterations that occur during tumor development. These lymphocytes, called neoantigen-specific, play an important role in the efficacy of immunotherapy. Given their ability to attack and eliminate tumors in a highly specific manner, the objective of the project is to develop personalized cell therapies based on the administration of large numbers of neoantigen-specific T lymphocytes. To this end, the presence of T lymphocytes that recognize mutated peptides derived from proteins that are recurrently mutated in cancer, such as KRAS, will be studied in cancer patients. Once identified, the T-cell receptor, responsible for conferring antitumor specificity to the lymphocyte, will be isolated. This receptor will be cloned and introduced into a viral vector to induce receptor expression in peripheral blood lymphocytes. In this way, we will generate an army of T lymphocytes capable of recognizing a specific tumor mutation.
